we got up at 5am today and got all bundled up to go skiing. then, cancelled. noooo! already wake up, pack lunch, etc then apparently there was heavy snow at the ski resort last night and it was flooded(??) so it was postponed. so we all trudged back in. then i realised i looked like a paper boy or something. i was wearing waterproof men's golf pants, possibly unisex jacket, unisex bag, and unisex beanie. haha.
kah wei called me this morning to tell me it was snowing. i looked out my window and wow it was so heavy! so i quickly called naz and got changed but by the time we went out, the snow had stopped. omg so cacat!! it snowed for like 5 minutes only!?! we could only see bits of ice on the grass. wth wth wth. how lame is that? sheesh
today was my last day of labs. what have i been doing?
my supervisor is really nice. she celebrated epiphany with us on january 6th. it's the 12th day of christmas when the 3 kings or wise men arrived in bethlehem. in france, where she's from, they celebrate by having a galette which traditionally contains a bean inside, and they cut the cake and whoever gets the bean is the king (or queen) of the day. the youngest person is supposed to not look while they cut the cake (in case the bean pops out) and then assign the pieces to everyone. i was the youngest so i had to turn around while she cut and then assign the pieces to people. i didn't get the bean but she had bought paper crowns for everyone from france so we each got one as a souvenir. she had even gotten into trouble at immigration for bringing the galette and the crowns. then we found out when her birthday was and got her a cake but besides that, we have been doing work. we did cell culture stuff to investigate the delivery of drugs to tumour cells. we prepared vesicles and complexes and tested them on cancer cell lines. some days we had to stay til 8pm but some days were short.
we seeded plates with cells in growth media (didn't take pictures of this but they're pink like the following picture, in plates like the second picture) and incubated them. then we sucked out the media and treated the cells with the vesicles or complexes or controls, and we incubated them again.
this is a series of dilutions for one treatment. the pink is from the medium. then we sucked out the treatment and analysed them with the mtt assay which colours living cells purple. we used columns 2-11. as you can see from this picture, the cells are dead in column 2 which has the highest concentration of treatment. we used micropipettes to fill all the wells with cells, but we only treated and analysed the wells we needed. a battery-operated glass pipette filler which is much easier to use than the bulbs and the pumps we had to use last time random huge 1000mL cylinder which we don't use but was in the lab
omg some people are so stupid. this guy dried his clothes in two dryers then when they were done, some clothes weren't dry yet so he took out some and left the rest in the two dryers. all the while, i was sitting there with my basket of wet clothes, obviously waiting for the dryers and he stillleft them in two dryers when the dryers are huge and have enough space for his clothes to fit into one. i had been nice enough to sit there for a few minutes waiting for him to come and collect his laundry. i should have just taken his clothes out and put mine in without waiting. sheesh.
omg my vista landlord is so unbelievably evil. until now he still never give back my deposit. he paid alvin and told him he'd pay me but never did. wth!!!!! i curse him to have bad luck for 20 years and go bankrupt. evil money-grabbing jerk and his wife!
don't rent vista b-3-7 coz the landlord is an asshole. in fact, go burn it down or smth. warn your friends if they live there. if i could remember his name, i'd put it here. mr lee smth. go die.
we went to the loch ness last thursday and stayed for two nights.
halfway up, the bus stopped for a toilet break and there was snow!!!!! at first the snow was really light, then it got heavier as we were going to board the bus. two happy people with snow in our hair :D mountains and sheep this is our hostel: loch ness backpackers it had a nice little fireplace with big sofas. there are books and games in the corner there and also a small spiderman tv with dvds to watch. the next day we headed for the loch ness. on the way, we stole snow from parked cars which had come from the mountains and threw snowballs at each other. haha. amir tried this on then he ended up buying this loch ness monster outside a hotel which serves really good vegetable soup another version of nessie we went on a boat ride by jacobite cruises to urquhart castle the trebuchet, which is a catapult the castle ruins i have a fear of falling down staircases so i don't like see-through stairs like this and narrow winding ones like these. naz laughed at me when i was going down so slowly. :P kah wei was caught taking a dump in a kiln then we waited at a hotel for a bus back to our hostel, but the bus left us behind so we ended up taking cabs back on the way home the next day, the bus stopped for a toilet break again and this time there was more snow i made this crude little snowman in like two minutes. so cute right?? hahaha. still hoping to make a proper big one. :)
